چکیده
Abstract Multi-analyte single cell technologies have potential to greatly accelerate antibody discovery by identifying antigen-specific B cells. Barcode Enabled Antigen Mapping (BEAM) provides a high-throughput, multimodal analysis of antigen-bound cells (BEAM-Ab) using 10x Genomics Chromium Single Cell Immune Profiling v2 Solution and novel computational approaches discover B-cell receptor (BCR) sequences for further functional characterization. To demonstrate the antigen sensitivity BEAM-Ab, we spiked in 5% Hen Egg Lysozyme (HEL) gp120 transgenic 90% wild type non-transgenic splenocytes. The were screened panel barcoded antigens then CD19+PE+ (antigen+) isolated flow cytometry. gene expression, BCR, barcode indicated clonotype specificity respective sorted Furthermore, demonstrated BEAM-Ab analyzing convalescent COVID patient sample against an containing five different negative control. Analysis this suggests clonal expansion recognizing ectodomain SARS-CoV2 spike protein, binding domain D614G mutant protein. We did not observe any Omicron-specific or non-specific control our dataset. Our data demonstrates that is effective tool. unlocks ability rapidly screen large numbers samples with accurate resolution specificity, entire workflow generating candidate just one week after processing.

Using high-throughput barcode sequencing to efficiently map connectomes
The function of a neural circuit is determined by the details of its synaptic connections. At present, the only available method for determining a neural wiring diagram with single synapse precision-a 'connectome'-is based on imaging methods that are slow, labor-intensive and expensive. Here, we present SYNseq, a method for converting the connectome into a form that can exploit the speed and lo...
متن کاملBarcode-enabled sequencing of plasmablast antibody repertoires in rheumatoid arthritis.
OBJECTIVE A hallmark of rheumatoid arthritis (RA) is the production of autoantibodies, including anti-citrullinated protein antibodies (ACPAs). Nevertheless, the specific targets of these autoantibodies remain incompletely defined. During an immune response, B cells specific for the inciting antigen(s) are activated and differentiate into plasmablasts, which are released into the blood. We unde...

the pharmaceutical ‘success story’ of the past decade. Mab-based pharmaceuticals command a worldwide market of more than $15 billion, and that figure will grow to exceed $30 billion by 2010. The proven safety and efficacy of Mabs as a drug class are driving faster developmental and regulatory timelines, and simplified routes from discovery to the clinic and from the clinic to the market. The dr...
متن کاملUsing Mendelian inheritance to improve high-throughput SNP discovery.
Restriction site-associated DNA sequencing or genotyping-by-sequencing (GBS) approaches allow for rapid and cost-effective discovery and genotyping of thousands of single-nucleotide polymorphisms (SNPs) in multiple individuals. However, rigorous quality control practices are needed to avoid high levels of error and bias with these reduced representation methods. We developed a formal statistica...
متن کاملHigh throughput production of mouse monoclonal antibodies using antigen microarrays.
Recent advances in proteomics research underscore the increasing need for high-affinity monoclonal antibodies, which are still generated with lengthy, low-throughput antibody production techniques. Here we present a semi-automated, high-throughput method of hybridoma generation and identification. Monoclonal antibodies were raised to different targets in single batch runs of 6-10 wk using multi...
